چکیده

The tridecapeptide neurotensine has been the subject of increased interest since its discovery and characterization in the brain1). Aspects of its pharmacology include physiological effects both in the periphery and the central nervous system2). In order to elucidate further its pathophysiological role, neurotensin agonists and antagonists are needed. In the course of our screening program to find substances that displace the neurotensin from its receptor, a receptor binding assay using guinea-pig brain membranes was performed according to GOEDERT et al.3). During this screening, RP 66453 was isolated as a novel secondary metabolite produced by an Actinomycetes strain. For the production of this compound the Streptomyces strain A 9738 was inoculated into a 2 liter Erlenmeyer flask containing 250ml of seed medium consisting of peptone 0.5%, yeast extract 0.5%, corn steep 0.5%, glucose 1.5%, CaCO3 0.3%, NaCl 0.5% and agar 0.1%.

Cytotoxicity effect of secondary metabolites of Streptomyces koyangensis and Streptomyces tunisiensis isolated from saline soils of Garmsar City on human breast cancer cell line (MCF-7, IBRC C10082)

Background: Streptomyces is one of the most important prokaryotic microorganisms, and their secondary metabolites have high antimicrobial and cytotoxic properties. Three-quarters of the antibiotics known to be produced by these bacteria. As a result, finding new effective compounds from these microorganisms can be a way to treat cancer. The aim of this study was to isolate and screen halophilic...

Extraction and Identification of Antibacterial Secondary Metabolites From Marine Streptomyces sp. VITBRK2

Actinomycetes were isolated from marine sediment samples collected from the east coast of Chennai, Tamil Nadu, India. Well diffusion and agar plug methods were used for the evaluation of antibiotic production by these isolates against drug resistant Methicillin- resistant Staphylococcus aureus (MRSA) and vancomycin resistant Enterococci (VRE). The potential isolate VITBRK2 was mass cultured for...

Oxoprothracarcin, a novel pyrrolo[1,4]benzodiazepine antibiotic from marine Streptomyces sp. M10946.

A novel pyrrolo[1,4]benzodiazepine antibiotic, designated oxoprothracarcin (3), was isolated from the marine strain Streptomyces sp. M10946 along with three known secondary metabolites, cyclo(D)-Pro-(D)-Val (1), cyclo(D)-Pro-(D)-Leu (2), and limazepine A (4). The chemical structures of these substances were elucidated by spectroscopic analyses, including 1D- and 2D-NMR and ESI-MS. Antitumor and...

A lyophilized peptide radiopharmaceutical kit to target neurotensin receptor for tumor imaging

Introduction: Neurotensin (NT) is a tridecapeptide that binds specifically to neurotensin receptors. Several forms of cancer, including small cell lung cancer, colon, pancreatic and prostate carcinomas especially exocrine pancreatic carcinomas express receptors for neurotensin peptide. Radiolabeled neurotensin derivatives with a high affinity for these receptors might be used for scintigraphy. ...
